I start out with one of the major independent sites, usually travelocity.com. That gives me an idea of the most I should have to spend, and some of the airlines that cover the route. Then I start looking in earnest.

orbitz.com is usually my next stop. The same tickets at orbitz are usually cheaper than travelocity, because orbitz is owned by the airlines. I'll also check the airlines own sites. Of course some airlines like Southwest only publish their fares on their own sites.

If the destination is a resort area (or if I'm starting from a resort area), I'll check with a travel agent. This is where they can really save you money, because the travel agents can offer you deals that you won't find anywhere else. It's not uncommon to get a round-trip flight from Detroit to Honolulu for 600 bucks, maybe with a rental car thrown in.

Also, don't forget to check nearby airports.
You can sometimes save a couple hundred bucks by driving a hundred miles.
