If that hole is at the top of the rear axle, ignore the 52 quarts for topping up or you may overflow all the seals. You do mean topping up, not refilling, don't you?
If that hole is low down the side, it is probably the level plug. If it is (check the service manual), fill until oil runs out the hole. Could be anything up to 52 quarts, depending on how low it is in the first place!
If you intend refilling the whole unit, you may find that there is more than one drain plug, if the lube is for the gearbox and differential - again, check the manual.
