You should plan to spend at least a few hours on the Blue Ridge Parkway. It's a very pleasant drive.
The North Carolina Zoological Part at Asheboro is good.
A nice coastal drive is to go from Wilmington, NC down to Fort Fisher, then take the ferry from Fort Fisher to Southport. The Fort Fisher aquarium, Fort Fisher, and Brunswick Town are all on this route.
A MUCH LONGER coastal drive is to take the ferry from Cedar Island, NC to Ocracoke, then drive up the Outer Banks to Nags Head. This is a full day trip. The ferry alone takes three hours. You can visit Cape Hatteras Lighthouse and Kitty Hawk.
One of my favorite places to visit is Tryon Palace in New Bern. The colonial palace is a reconstruction, but it is furnished in real antiques from the period. The gardens on the grounds are impressive as well.
