The missing tooth designates that particular cylinder is TDC on compression stroke ( cam/crank synchronization). A crankshaft position sensor can tell you TDC but not if its on compression stroke or 180* out. The cam sensor info is mainly used to tell EFI when to inject fuel in firing order to specific cylinders on sequential/port fuel injection, not really needed on bank/batch fire, single injector or TBI injection systems.
