There is a drain plug in the bottom of the carb that is packed with some fine material that lets gas drain out but will not let a flame pass. If the hole is open it would suck dirty air into the engine. Leaving the intake pipe off may reduce vacuum slightly but not enough you would notice a difference in performance.
