Only thing I can think of is that maybe there is something not letting the plug ground out there,dirt or grease or something.I agree with Andy that 1 cylider should make too big of a difference in the power,might want ot recheck the timing.Are you 100% sure the new manifold is sealed up tight?When you pulled the wire off and got the spark,did you go to the plug or just a spot on the engine?Try going to the plug and see what happens.
