There is a Two Stroke Detroit Diesel group over on yahoo.com. Those guys pretty well know every trick in the book.
Compression ratio, cam timing, turbo size and exhaust temp control are all critical. Water injection downstream of the turbo's and perhapes even intercooling will be vital.
A silver series 6V53TI would be a good base engine to start with.
