It probably varies from state to state, but I live in Nebraska and I've bought a number of vehicles in South Dakota.
Here, I need to take the SD (or other state) title to the local Sheriff's office where they run the VIN to make sure it isn't stolen or have anything else dire in it's history. Then I simply take the form from the Sheriff's office, along with the SD title, to the County Clerk's office and follow the usual drill to get a Nebraska title and registration.
Each state has its own procedures, so it might be time well spent to visit the local office where you register vehicles to find out beforehand.
