No, with the tube plugged the level can't move unless you get the air in to move just enough to show the true level.

Opening the drain is a little different than using a tube. With the drain open some gas will always run out and air will replace it. But I've never personally used a tube that way.

Go ahead and pull the carb bottom. It is easy and good practice. Don't drop the venturi on a hard surface. It is in the carb throat and sometimes falls out after disassembly. There will be something to clean out for your effort.
