On some tractors the sound of a leaking valve, and associated misfire can be detected only at certain speeds. A leakdown check would detect the issue, but requires removal of the injectors. A compression check is the same. I would adjust the valves (looking for a tight clearance on one valve.) I would also crack the injector lines one at a time to find the misfire (assumes it does it in neutral sitting in the yard) Exhaust manifold temperature analysis (with a infrared hand held) might also be decisive in locating which cylinder is missing.
