Sorry, misread your 1st sentence where you said you used carb cleaner -- I would soak it again. You can tell whether most of the passages are clean by seeing if you get air out somewhere when blowing it out. I would go easy with the manual cleaning. The problem with the lack of response to the main adjustment is a common one. A new needle and seat should take care of it, provided the passage is clean, IF you can get the old seat out.
