I work for an ISP. Outlook is known for corrupting profiles. If your ISP isn't having issues. You can try reentering the SMTP server name.
Next I would try deleting the profile in Outlook and setting up a completely new one. Write down your current settings: SMTP server, POP3 server. ect. Delete the old profile by going to Tools->Accounts-> be on the mail tab, highlight and remove the profile. Then click add-> mail and run through the setup to make the new profile. This doesn't delete any mailboxes so no mail will be lost it just resets the profile with all the server info. If it still won't send it maybe a corrupted outbox.
