Assuming it is an insulating board I would just stuff as much fiberglass insulation into the hole so that the cavity is full below the hole. Then inject expanding foam into the hole and let some of it expand out of the hole. Once dry shave it off flush with a butcher knife. From there I would put some kind of metal flashing over the area before you put the siding back. That would prevent any varmit from coming through again.
