3010 Hydraulic Fluid Level - Check Running or Not

Do you check the hydraulic fluid level with the tractor running or not, hot or cold, cylinders on loader extended or retracted?

I cleaned the screen and changed the hydraulic filter and filled with genuine JD fluid to the max line on dipstick with it NOT running and resting on the threads on level ground but it didn't take near the amount of fluid speced in the operators manual, even after running and shutting off and checking.

You did it correctly. The position of the cylindrs don't matter unless that are single acting.
 
The oil level should be checked with the dip stick resting on the threads. If you checked it by screwing it down, you will get an incorrect reading. (engine is the same as well)

Then I suppose for correct level the 3pt should be down. I always check mine with 3pt up because it stays up.
