No, it depends on the primer -- look at the instructions. If none, get another brand/type. Two part epoxy is the best primer and is not sandable. On surfaces you want smooth, such as sheet metal, you put a surfacer over the epoxy and sand it. Do not use the surfacer as the main primer, surfacers/primer surfacers do not adhere to the metal as well as epoxy. On cast and other non critical parts as far as smoothness, just used epoxy without surfacer.
 
It may not always be necessary to sand all the primer however the paint job would be better if you did. It's always best to scuff a finish prior to putting on any coating. Then if the primer you are using is epoxy and you have exceeded the recoat window it really is necessary to scuff the finish or put a fresh coat of primer on.
 
As Stephen mentioned, epoxy outside the window has to be sanded or scuffed AND a new coat put on. It is not supposed to be sanded within the window, usually 3-7 days at 70 degrees -- read the instructions to find the window.
