My 1955 Ford 640 has the block off plate with a short pipe nipple sticking out of it with a plug on the end. What would I need to do to operate a one way cylinder from this line? Would it compromise the 3 point lift?

You can either use it for the cylinder or the 3 point but not both at the same time because you have to lock the lift arms in the down position for it to be able to work the cylinder. Only way to have both working is to buy the proper aux hyd set up

Also you may want to consider putting in an open/close valve in so that if you want to use the 3pt you don't have to undo and plug whatever you have the cylinder mounted to. A quick disconnect adapter can also be used disconnect the line when not needed. Here's one of my set ups on my 8n.

as has been said, chain 3 pt, and then you can run a sa cyl using the touch control.

as Kirk said. add in a valve, so you can then unchain and use 3pt.

You can get a push / pull diverter valve to prevent chaining and external valving, or a more expensive true bolt on remote.
