1970 3000 GAS MISSING ON one cylinder?

I recently aquired a ford 3000 gas,8 speed on a trade. I converted the horizontal exhaust to a vertical exhaust and noticed tonight it has a miss. I determined its the middle cylinder that isnt firing by pulling off the plug wire and no change in engine.. i swapped the plug and wire off another 3000 i have and it didnt make a difference. plug has spark. i checked points and cleaned them and they look alright. the oil and antifreeze both look good, no smoke at all out of exhaust, and no bubbles in radiator. i did not check compression yet but when i took the plug out i cranked it over and it felt and sounded like it had decent compression just with my hand over the hole. I am stumped and hope its nothing serious!! Any ideas or suggestions much appreciated! -John
 
Check the compression.
May be a bent pushrod or stuck valve. If a valve isn't
opening, or only opening part way, it could cause a
miss.
 

Check the compression, it takes a min of 100 psi for a cylinder to operate properly. If compression is good check for a leak where the manifold mates to the head. If compression is low pull the valve cover an check for bent push rod, broken valve spring, valve lash adjustment.
 
agreed with the others, if that hole has spark, then do a proper comp test on it. just blowing some air tells you a little, but not enough.
