I have a Ford 5000 diesel and it is progressively getting slower to engage in reverse, it can take over a minute to go. Its worse when cold. I was told to use regular ATF fluid and I have added some over last year due to leak, not sure if this could be causing issue since now I'm reading shop manual and it calls for M2C41-A transmission and hydraulic oil ( what ever that is) or if it needs some kind of adjustment or fluid/filter change. Most forward gears seem fine but it does seem to slip some under some load conditions. Any ideas or recommendations??

It is not uncommon for late model S-O-S transmissions to delay upon reverse engagement but a full minute is excessive.
You should use only CNH 134D oil or quality equivalent. Do not use ATF.
When did you last change the filter?

Try a band adjustment, as well as changing oil and filters. Band 3 is likely your culprit. But as Dean says, ATF is a no no! 134 D or equivalent. It supersedes M2C41.
I have not serviced the fluid yet, I purchased the tractor about 2 years ago from the original owner and followed his advice to use ATF. Once I started having issues I purchased a service manual and read about the fluid and also adjusting the bands. I will start with the fluid and go from there. I was wanting so expert advice so thank you! It's a great tractor so I was hoping I could fix it.
I also have a 1967 5000D SOS and it has had the same problem since the day I bought it. Have you tried 9th and 10th gear also? I'd be willing to bet you that they are also slow to engage. It's Clutch 1 that is common to R1, R2, 9, 10.

Reading the manual can instill some apprehension , but it's pretty straight forward taken one step at a time. I would suggest draining all fluid from the trans. Clean the pickup screen and install a new filter and fresh UTF fluid that meets ford specs. Warm tractor up to operating temp and adjust bands following manual precisely. As Larry says, it sounds like band three, but adjust them all in the correct sequence. You should then be good to go.
