runs fine cold , has fresh air and fuel filters. after warmed up and put under a load it starts missing on one cylinder just started doing it
 
You should have 65 lbs fuel pressure when wrapped up. Most times it is a fuel flow problem on a Detroit. They use a high flow system. 80% of the fuel flow in them is used to cool the injectors. If you have the needle style injector I wouldnt be suprised if it is getting sticky when hot. I dont think this would be an issue on the crown valve type. The tag will start with N or C , thats how you know wich you have. It wouldnt hurt to have your injectors checked. If there isnt a shop in your area that tests old detroit unit injectors give me a call I have a test bench for those. 231/757/0064
 
Each cylinder has its own injector and its own injector pump. Chances are, if just one cylinder skips and the others do not, that's what I'd check.
Since the injectors/pumps are activated by mechanical rocker arms - check the adjustment on them. My 3-53 does sort of the converse. It's had a cold skip for 20 years. Only runs on two cylinders until it warms up and is then fine. Since mine runs good hot, I've never bothered to pull the valve-cover off to adjust the rocker-rack.
