I have never used a sealer on a manifold gasket myself. I would thin the most important thing is to make sure that the sealing surfaces are as clean as humanly possible.

In the past, I have used what I call aircraft schelac, only on the intake portion, of the manifold, because I couldn't get it smooth enough to suit me. A man shouldn't need any sealer if it is clean enough.
 
More important than sealer is having the manifold and head surfaced so you have good flat surfaces to seal on. The time to do it is when everything is apart.
